For a long time, the world of genetics was forced to choose between two compromises: either highly accurate but short data fragments or long but "noisy," error-prone sequences. Pacific Biosciences (PacBio) has solved this equation by creating HiFi (High Fidelity) technology.
Today, it is not just another sequencing method – it is a tool that allows you to see the genome as it truly is: integral, precise, and multifaceted.
The Heart of the Technology: SMRT Sequencing and the Magic of ZMW
PacBio's uniqueness begins at the nanoscale. It is based on SMRT (Single Molecule, Real-Time) technology – sequencing individual molecules in real-time.
How does it work? Imagine the surface of a chip (SMRT Cell) containing millions of microscopic wells called ZMWs (Zero-Mode Waveguides).
- Nano-volume: The diameter of each well is so small that light cannot fully penetrate inside – it illuminates only the very bottom layer where a single DNA polymerase molecule is anchored.
- Light Code: As the polymerase incorporates a nucleotide (labeled with a fluorophore), the system instantly detects a flash of a specific color. This happens thousands of times per second in millions of wells simultaneously.
This allows for the "live" observation of the enzyme's activity, obtaining primary data directly from the native DNA molecule.
HiFi Reads: Where Length Meets Flawless Accuracy
Unlike competing solutions where long reads have a high error rate, PacBio HiFi technology ensures impeccable accuracy. This is made possible by a brilliant engineering feat – Circular Consensus Sequencing (CCS).
The DNA is formed into a circular structure (SMRTbell), allowing the polymerase to move along a closed loop multiple times. The system reads the same molecule dozens of times, after which the software compares these "laps." Random errors are neutralized, resulting in a HiFi read – a sequence up to 20,000 base pairs long with 99.9% accuracy (Q30 and higher). This enables the assembly of complex genomic regions that were previously considered "dark matter."
Why Your Laboratory Needs PacBio
Conquering "Dark Regions"
The human genome is not homogeneous; it is saturated with repeats, palindromes, and regions with extreme GC content. Other technologies often get "lost" in these areas or make systematic errors. PacBio, by eliminating the PCR amplification step, traverses these regions with ideal coverage uniformity.
Direct Methylation Detection
One of the most impressive advantages of PacBio is the ability to study DNA methylation simultaneously with primary sequencing, without any additional sample preparation.
Why is this important? Traditional methods require bisulfite modification, which is aggressive, damages the DNA, and significantly increases costs. PacBio technology works differently:
- The instrument measures the kinetics (speed) of nucleotide incorporation.
- When the polymerase encounters a methylated base, it changes its pace for milliseconds.
- These delays are captured by the system, providing a complete methylation map of the entire genome.
You receive data on the native state of the DNA without chemical damage or additional reagent costs.
SPRQ-Nx and Multi-use SMRT Cells
The latest SPRQ-Nx chemistry makes HiFi sequencing not only accurate but also highly cost-effective for laboratory budgets.
PacBio’s unique development allows a single SMRT Cell to be used for sequencing up to three times! This radically reduces the cost per study and ensures maximum optimization of reagent expenses.
As a result, the cost of a high-precision HiFi genome is now approaching the price of standard short-read sequencing. At the same time, the value of HiFi data is significantly higher: it does not require complex de novo assembly or expensive verification by alternative methods.
Areas Where PacBio Remains Unrivaled
- De novo Genome Assembly: Creating reference genomes for new species of plants, animals, or pathogens with zero errors.
- Medical Genetics: Detection of structural variations (inversions, translocations) that cause rare genetic diseases but are too large for short-read technologies.
- Full-length RNA Sequencing (Iso-Seq): View complete transcript isoforms from the 5' to the 3' end rather than fragmented segments. This fundamentally changes our understanding of gene expression.
Flagship PacBio Solutions: Unlock the Power of Revio and Vega
PacBio offers a versatile range of equipment designed to scale with your needs, from small-scale laboratories to high-throughput genomic centers.
PacBio Revio: A New Era of High-Throughput HiFi Sequencing
Revio is a technological breakthrough designed to make HiFi sequencing the primary method for Whole Genome Sequencing (WGS).
- Incredible Throughput: A single instrument can sequence up to 1,300 human genomes per year.
- On-board Processing: Previously, converting primary signals into HiFi reads required massive external computing clusters. Revio performs this internally using AI (deep learning algorithms) to recognize DNA bases in real-time.
- Google Health DeepConsensus: PacBio officially collaborates with Google, integrating their DeepConsensus deep learning models directly into Revio. This allows the instrument to not just "read" but intelligently correct errors "on the fly," ensuring 99.9% accuracy.
- Cost-Efficiency: By optimizing processes, Revio lowers the cost per genome, making long reads more accessible than ever.
